Gene Expression Core Facility
Purpose
Studies of the effects of toxicants on gene expression are integral to mechanistic toxicology investigations. The ability to analyze gene expression at multiple levels is important for the research in MCTEH. This core contains state-of-the-art equipment necessary to successfully study a broad spectrum of gene expression at the RNA and protein levels.
Services
The Gene Expression Core Facility currently assists investigators with experimental design, sample preparation and analysis of gene expression as a collaborative effort. To use the Core, the Principal Investigator sends a technician, graduate student or post-doctoral fellow to be trained in the application of a particular assay. After training has been completed, that individual is then equipped to perform the assays for the project using Core equipment.
Management
The Gene Expression Core Facility is developing and Dr. John Wise is currently its Director. The Wise Laboratory’s portfolio of funded projects makes extensive use of microarrays and quantitative PCR. Dr. Wise has also maintained close ties with Yale’s Microarray Core Facility. Consequently, Dr. Wise is responsible for the equipment within the Core and is overseeing the training and supervision of technical staff and students on those instruments.
Equipment
This Core Facility already contains some equipment needed for gene expression studies including: A DNA sequencer, a LiCor Odyssey infrared imager for imaging gene expression; a spectrophotometer for quantitating nucleic acids and protein; a phosphoimager for imaging gene expression with radionuclides; an Agilent bioanalyzer for assessing quality of nucleic acids and protein, a -80°C freezer for storing samples; and a high speed centrifuge for large scale extractions. The Wise Laboratory also has an Axon GenePix 4200A 4 color microarray scanner and a Stratagene MX4000 quantitative PCR machine that are used commonly by other investigators in the Center.
